Applications Used in Both Study The studied samples were subjected to the following: Bioinformatics: 1-Sequence analysis and selection of PCR primers using gene bank and software. 2- DNA Synthesis, primer (Oligonucleotide) synthesis and purification. 3- Measurement of oligonucleotide concentration. 4- Extraction of nucleic acids from eye swabs by 6 different methods for evaluations. 5- End point amplification PCR amplification (Quantitative PCR by gel documentation system). 6- Detection of the amplified product and DNA analysis by agarose gel electrophoresis. 7-Real-time PCR. 8-Real-time NASBA 9-Primers and internal controls for the previous amplification methods were synthesized by DNA synthesizer, site directed mutagenesis and cloning technique. Cloning, DNA isolation from plasmid vectors (positive control).
